MAJOR TIMOTHY CAIN

Dr Isaacs was in charge of developing the Nemesis Project but had to answer to Cain, who was his superior and in charge of Isaacs's research. However, Isaacs loathed the man with a passion, not just because he was his boss but moreso because he considered the man a complete imbecile.
Isaacs was a very thorough man of science who liked to carry out his work meticulously which of course clashed considerably with Cain's more impatient and inattentive approach.
Isaacs believed that Cain was singlehandedly responsible for the Raccoon disaster (and thereafter the end of the world) who had decided to go against Isaccs's recommendation of sealing the Hive with concrete.

ALICE ABERNATHY

Isaacs looked upon Alice as his "pet project". His research for the Nemesis Project had been going nowhere until finally he'd been given his perfect test subject - Alice.
Her ability to adapt with the T-virus and take it to a whole new level fascinated him and he considered her his greatest success. He had feared that the Nemesis Project had been compromised beyond all recognition but after salvaging her corpse from the helicopter wreck he'd been able to fully restore her and was overjoyed that he could finally move forward.
He was in awe of her and knew that if she remembered who she was he wouldn't be able to contain her and so instead let her believe she was free.
